Angiogenesis is the process by which a new blood supply is established from pre-existing blood vessels. The process is initiated by degradation of the vessel basement membrane, endothelial cell (EC) proliferation, invasion, directional migration towards chemoattractants, tube formation, and finally the establishment of new vasculature. In this workshop we will discuss a portfolio of BD products designed to investigate specific stages of through standardized and quantitative in vitro cell-based assays. Development of new angiogenesis modulating therapeutics has been hampered by the lack of suitable cell-based assays. The BD BioCoat assay platforms, directed at multiple aspects of angiogenesis, address this critical need in drug development and basic research. This assay platform, with automated imaging on the Pathway Bioimager facilitates scale up for screening purposes. We will demonstrate their use in 3-D endothelial cell tube formation analyzed in confocal mode on the Pathway in place at the ATC. During the workshop we will capture images on the system, as well as discuss the application of this instrument to real-time analysis for live cell assays. Software tools to analyze the data and provide quantitative result will be presented.
