Description:

Objective:

The overall objective of the Southern California Particle Center and Supersite (SCPCS) is to bring together outstanding scientists from the leading universities in Southern California to identify and conduct high priority research to better understand the effects of particulate matter (PM) and ensure protection of public health. The SCPCS makes use of an integrated approach to address each of the areas of Exposure, Dosimetry, Toxicology, and Epidemiology. This integration is accomplished by seeking out and involving in the Center some of the major figures in air quality and environmental health. A team has been assembled for SCPCS that is committed to the need for strong cross- and interdisciplinary programs of research in order to address the challenging issues posed by PM. By improving our fundamental and observational understanding of the complex relation between particle exposure and human health, it is our goal to lay a firm scientific foundation for effective intervention strategies for public health protection.

The Center will conduct research that addresses the objectives listed in the March, 1998 NRC report entitled Research Priorities for Airborne Particulate Matter: 1. Immediate Priorities and Long-Range Research Portfolio. The projects of this Center are based on the ten research priorities outlined in the NRC report. Related research objectives of the SCPCS include: To make use of ambient particle concentrators for exposure assessment and toxicological research; To emphasize chemical and physical characterization of PM, especially size distribution, in the study of the relationship between PM and associated morbidity and mortality; To emphasize mechanistic toxicological research including in vitro, animal and human approaches for better understanding of the underlying bases for increased morbidity and mortality associated with exposure to PM; To conduct toxicologic research which reduces the uncertainties in the identification of causitive PM constituents; To investigate the role of allergens and bioaerosols in the morbidity and mortality associated with PM exposure; to better characterize the relationship between mobile sources and airborne PM in the Southern California air basin; and, To investigate the relation between outdoor, indoor and personal exposure to PM.

Approach:

The SCPCS addresses each of four disciplinary areas central to understanding the complex particulate matter issue. These areas of Exposure, Toxicology, and Epidemiology form the Research Cores of SCPCS. To assist the Research Cores we have established four support Cores. The Biostatistics Core will provide statistical support for research projects conducted by Center investigators and to investigate design and statistical analysis issues related to the Center research studies. The Concentrator Exposure Facility is designed to conduct inhalation studies to ambient course, fine and ultrafine particles. This core provides a unique transportable exposure facility capable of concentrating ambient particles for use in toxicologic tests with laboratory animals and clinical exposures with humans. Recent technology developments allow for the concentration of real ambient particles rather than generation of synthetic or re-suspended aerosols for toxicological studies. This opens the possibility of conducting experiments over a full range of realistic concentrations and for particulate matter levels found in episode conditions. The Analytical Chemistry Core is to provide high quality, state of the art, analytical chemistry services as well as field and laboratory exposure characterization capability to Center investigators. This service core integrates the analytical and field sampling resources on the UCLA campus into a well defined and organized structure. The Dosimetry Core will be considered both a research and support core. This core will support Center investigators with dose calculations for Exposure, Toxicology, and Epidemiology projects. The core will also initiate original research in collaboration with other investigators to improve the design and interpretation of studies as well as to improve dosimetry models.

Southern California also has some important resources that reinforce the Center's focus and leverages EPA's investment in a Los Angeles-based center. These resources include an NIEHS Environmental Health Sciences Center, a joint EPA/NIEHS Children's Health Center, an excellent network of PM monitors sponsored or operated by the California Air Resources Board (ARB), EPA, and the South Coast Air Quality Management District (SCAQMD).

An Administrative Core will coordinate the multi-campus research effort, provide for internal program review, and work to ensure that the team functions effectively as a center by encouraging and maximizing cross campus and interdisciplinary activities. An Internal Advisory Committee will be established to ensure that effective interaction across Cores is occurring, to identify new research priorities and projects, to evaluate the overall research productivity, to determine appropriate commitments to Center research activities, and to insure young investigators are being encouraged to participate in the Center.

The approach of SCPCS is to seek out projects for support under the Research Cores that address one or more of the top ten research priorities for PM that were identified in the March, 1998 NRC Research Priorities report. We have assembled a strong program of research that addresses the NRC priorities. The SCPCS is comprised of nine initial projects within the Research Cores. Specific SCPCS projects in the Exposure Core focus on aerosol characterization, personal exposure, and exposure modeling.

Activities under the aerosol characterization project include the development of a mobile Particle Instrumentation Unit (PIU) that will be used for mechanistic aerosol field studies as well as studies in this and other Research Cores that require state-of-art PM measurements. Other studies within this area include characterization of ambient ultrafine aerosols prior to planned toxicologic studies. Characterization studies will also be conducted for aerosol constituents such as PAH compounds, metals, and oxidants. Within the aerosol characterization project area we will evaluate sources and concentrations of aero-allergens in Southern California.

In the personal exposure project area we will conduct personal and residential measurement of aerosol exposure and concentration in schoolchildren (sensitive population) participating in an ongoing study in the Epidemiology Core. Integrated particle samples will be collected in three size ranges: 0-1, 1-2.5 and 2.5-10m. Real-time variation will be monitored with nephelometers. Data obtained in this study will be used to validate probabilistic population exposure models for PM.

The exposure modeling studies will connect sources of PM constituents with regional air quality and human exposures. The modeling efforts will be fundamental, involving chemical kinetics and mechanisms and will support exposure studies in this and other Research Cores. Modeling efforts will include trajectory aerosol modeling, regional exposure modeling, the SMOG air quality model and the REHEX Regional Exposure Model.

The Toxicology Research Core initially involves four projects. The aim of the projects in this research core focus on: 1) identification of physical and chemical components of PM that generate or enhance the following pathological responses: IgE production, airway inflammation, airway hyper-reactivity, bronchial hyper-reactivity, cardiac and systemic coagulation abnormalities, production of reactive oxygen species and DNA adducts., and 2) determination of the biological mechanisms whereby the identified responsible components of PM elicit the above pathological responses.

In the asthma exacerbation study a mouse model will be used to test the hypothesis that PM enhances antigen-specific IgE response. Further testing will be conducted to identify mechanisms and components of PM responsible for exacerbation of asthma response. This project will test the hypothesis that airborne particles enhance antigen-specific IgE response in mice, and seeks to identify the responsible physical and chemical constituents causing this effect.

In the allergic inflammation study in-vitro cell cultures and the asthma mouse model will be used to examine adjuvant effects of PAH and redox-active quinones in PM on allergic inflammation. The hypothesis in this study is that PAH compounds induce P4501A1 (CYP1A1) expression through interaction with the AHR in cell types such as macrophages and epithelial cells. Specific objectives are to: 1) establish relevant models for OVA-induced asthma in mice to determine whether concomitant exposure to PAH and redox-active quinones will enhance allergic inflammation and airway hyperreactivity, and 2) conduct in vitro cell culture experiments to determine the effects of PAH and redox active quinones on the induction of cytokine and chemokine messages in pulmonary alveolar macrophages, epithelial cells and their cell lines.

In the airborne PM and quinone study, quinone reaction with nucleophiles will be examined for adduct formation that may act as antigens and chemical carcinogens. The hypothesis to be tested is that PM contain quinones that react with tissue nucleophiles to form adducts that act as antigens, catalysts for reactive oxygen mediated toxicity and chemical carcinogens. This project will be linked with the epidemiology core in the third year of the Center to assess PM in areas with known high incidence of asthma.

In the human clinical exposure study, chamber studies will be conducted on human volunteers to evaluate acute cardiopulmonary responses to concentrated ambient ultrafine, fine and coarse PM. Testing will be conducted on subjects with COPD following exposure to concentrated ambient aerosols. The hypothesis to be tested in this study is that controlled exposure to concentrated particles elicit statistically or clinically significant acute pulmonary or systemic responses in human volunteers. This hypothesis will be tested in multiple experiments involving different particle size ranges and susceptible human population.

The Epidemiology Research Core builds on an already established respiratory disease core of the NIEHS supported Environmental Health Sciences Center. The new activities of the SCPCS Center serve to strengthen an ongoing major inquiry into the chronic health effects of air pollution in Southern California children supported by the California Air Resources Board. In that study about 4000 school children living in twelve communities with differing levels and types of air pollution have been under study since 1993. While interesting findings have emerged from this study suggesting a relationship between pollution levels and health effects, there are still many questions that can only be addressed through more careful examination of dosimetry. The SCPCS supports two new projects initiated to address issues of key concern in this study. The mobile source study estimates exposure to and examines the influence of mobile source emissions on respiratory effects in the participating school children using information on traffic density and proximity of roadways. The bioaerosol study tests for the respiratory health effects of measured bioaerosols (including pollens, spores and fungi) within the same epidemiologic study.

Expected Results:

The Los Angeles Basin is home to more than 14 million individuals who breathe the most polluted air in the nation. The Research Core projects in this Center application were selected with attention to the public health objectives of the RFA and the ten major NRC objectives. Through projects described in this application the airshed will receive detailed characterization by exposure studies to assess PM size, composition, and spatial variability. Toxicology investigations will be directed toward a fundamental understanding of the mechanisms and the chemical and physical components of PM. The epidemiologic research on bioaerosols and vehicular exposure are likely to provide new information to enhance our ability to detect human health effects from ambient air pollution. New epidemiologic studies will be designed to begin in year 3 to apply the toxicologic research and exposure characterization to investigations of sensitive populations, particularly children and asthmatics. Novel studies will be conducted on human activities and allergens in airborne particulate matter, and their role in respiratory disease. The relationships will be quantified between indoor and outdoor particle levels and personal exposure to particulate matter. This will be conducted for schoolchildren participating in a major epidemiology study within the Los Angeles Air Basin (LAB). A coordinated modeling and analysis program will link aerosol sources and monitoring data with particulate properties and human exposure, from source/receptor relationships to regional scale simulations for the LAB, which will provide a more substantial basis for interpreting and applying experimental data, and in designing regulatory actions. Toxicology experiments will address exacerbation of the asthmatic response in a mouse model, stimulation of production of reactive oxygen species and DNA adducts in a rat model, and acute elicitation of pathological pulmonary and cardiac responses in potentially susceptible human populations. Complementary studies will seek to define the mechanisms and pathways whereby PM and the relevant causative constituents of PM elicit the above pathological responses. Successful completion of the toxicology studies will provide information that is essential for the future design of methodologies for identifying susceptible human populations and for preventing or ameliorating the deleterious effects of PM on the general population.
