Better Microbes,
Better Enzymes

Biomass Deconstruction and Conversion research is centered on consolidated bioprocessing (CBP), in which the same microorganism deconstructs and ferments biomass to yield the desired products. If the BioEnergy Science Center (BESC) can overcome biomass recalcitrance through CBP, it will reduce the need for pretreatment chemicals that are toxic to microbes as well as decrease the energy required to deconstruct and convert biomass into biofuels.

Two key hypotheses drive this BESC research effort:

  • Microorganisms can be engineered to enable CBP.
  • Enzymes and microbial biocatalysts can be understood and engineered to synergize with recalcitrance-reducing plant modifications to achieve better biomass deconstruction.

To test these hypotheses, BESC targets three microbial CBP platforms: yeast, Clostridium thermocellum, and Caldicellulosiruptor species. In yeast, a robust industrial ethanol producer, researchers have pursued genome modifications aimed at improving the expression and activity of enzymes that will digest cellulose. Already natural cellulose degraders, the other two microbes are thermophilic anaerobes whose metabolism is being manipulated by BESC to produce useful products such as ethanol or butanol. BESC is also demonstrating the synergistic action of improved CBP on modified plant cells.

Studies of microbial cellulose utilization at all levels—including enzyme components, enzyme systems, and pure and mixed cultures—show the increased effectiveness of microbial cultures compared with free enzymes acting in the absence of cells. BESC has extended this CBP proof of concept by genetically engineering C. thermocellum to make isobutanol from cellulose.

Dr. Lee Lynd is a professor of engineering and biological sciences at Dartmouth College. He is a recipient of the National Science Foundation's Presidential Young Investigator Award, as well as a two-time recipient of the Charles A. Lindbergh Award for his efforts to promote balance between technological progress and preservation of the natural and human environments. He has received the Charles D. Scott Award for outstanding contributions to the field of biotechnology for fuels and chemicals and is the inaugural winner of the Lemelson-Massachusetts Institute of Technology Sustainability Award.
