The DOE Joint Genome Institute will provide a 3x draft sequence of the female Populus trichocarpa clone ‘Nisqually-1’ in late-2002 and a second 3x draft in early to mid-2003. A whole-genome shotgun approach is generally planned to sequence the Populus genome. Genomic DNA will be fragmented into pieces of defined size, cloned into a plasmid vector, amplified, and sequence reads generated from a large number of sub-clones. Sequence reads will be assembled into larger contiguous regions and these further assembled onto ca. 2000 BAC-end sequenced scaffolds. Final assembly will be accomplished using supplemental information from physical and genetic maps of the Populus genome. All sequence data will be freely available to the scientific community.
