Air Quality Forecast Guidance

NOAA's National Weather Service National Air Quality Forecast System (AQFS) Graphic Forecast displays (http://www.weather.gov/aq) are web based presentations of gridded forecast guidance originating from the National Centers for Environmental Prediction (NCEP). Ozone and smoke data are displayed for a domain covering contiguous United States, Alaska and Hawaii. Ozone is shown as 1-hour and 8-hour average concentrations(in parts per billion or ppb), and smoke concentration in 1-hour averages, updated twice daily. Fire locations are provided by the NOAA's Satellite and Information Service Hazard Mapping System. Additional information on air quailty can be found under EPA's AIRNow site. The Toxics Release Inventory (TRI) Chemical Hazard Information Profiles (TRI-CHIP) dataset contains hazard information about the chemicals reported in TRI. Users can use this XML-format dataset to create their own databases and hazard analyses of TRI chemicals. The hazard information is compiled from a series of authoritative sources including the Integrated Risk Information System (IRIS). The dataset is provided as a downloadable .zip file that when extracted provides XML files and schemas for the hazard information tables.

ECOTOX is the largest database compilation of ecotoxicity information in the world, providing data on the adverse effects of single chemical stressors to ecologically relevant aquatic and terrestrial species. The primary source of ECOTOX data is the peer-reviewed literature, resulting from comprehensive searches of the open literature followed by abstraction and review of data that meet minimum data quality requirements. ECOTOX also includes third-party data collections from U.S. EPA Program Offices, the U.S. Geologic Survey, Russia, and OECD member nations. Data are not interpreted during the coding process, representing the information on species, chemicals, test methods and toxicity results as presented by the author(s).

Background: Chemical toxicity testing is being transformed by advances in biology and computer modeling, concerns over animal use and the thousands of environmental chemicals lacking toxicity data. EPA's ToxCast program aims to address these concerns by screening and prioritizing chemicals for potential human toxicity using in vitro assays and in silico approaches. Objectives: This project aims to evaluate the use of in vitro assays for understanding the types of molecular and pathway perturbations caused by environmental chemicals and to build initial prioritization models of in vivo toxicity. Methods: We tested 309 mostly pesticide active chemicals in 467 assays across 9 technologies, including high-throughput cell-free assays and cell-based assays in multiple human primary cells and cell lines, plus rat primary hepatocytes. Both individual and composite scores for effects on genes and pathways were analyzed. Results: Chemicals display a broad spectrum of activity at the molecular and pathway levels. Many expected interactions are seen, including endocrine and xenobiotic metabolism enzyme activity. Chemicals range in promiscuity across pathways, from no activity to affecting dozens of pathways. We find a statistically significant inverse association between the number of pathways perturbed by a chemical at low in vitro concentrations and the lowest in vivo dose at which a chemical causes toxicity. We also find associations between a small set in vitro assays and rodent liver lesion formation. Conclusions: This approach promises to provide meaningful data on the thousands of untested environmental chemicals, and to guide targeted testing of environmental contaminants.
