Abstract : BACKGROUND

Research on the Flavobacterial pathogens of fishes e.g. F. psychrophilum has been hampered by their inherent difficulties of primary isolation from fish, growth, and storage and recovery. Fungal overgrowth on primary isolation plates is also a major problem. There is a need for an improved isolation and growth medium, especially for (early) primary isolation from carrier, asymptomatic fish. Recently, we formulated a medium to recover F. psychrophilum isolates from long-duration frozen storage. We recovered 93+ % of about 150 isolates; this compared with 0 % in a previous attempt with the medium that most diagnosticians and researchers have historically employed. Arguably, recovery of F. psychrophilum from frozen storage is more challenging than primary isolation from diseased fish. We propose to develop this medium, to include contaminant deterrents, for use for primary isolation of F. psychrophilum. Three of the investigators (besides the PI) are members of the Great Lakes Fish Health Committee.

OBJECTIVES

1) Provide an improved medium for primary isolation and culture of F. psychrophilum;

2) Improve detection of F. psychrophilum from subclinically infected fish (screening tool; and

3) Ultimately, the medium will be recommended for use by groups/publications such as the AFS-FHS Blue Book, the joint AFS-USFWS Inspection Procedures Manual, the USFWS Wild Fish Health Survey Manual and the Canadian Fish Health Protection Regulations “Manual of Compliance”.
