Description: | Dario Vignali is interested in the regulation of T cell tolerance. In order to study how signaling by the TCR controls T cell responses in vivo he has developed an efficient and rapid system to generate “retrogenic” mice, which are derived from stem cells transduced with retrovirus vectors. By this approach he has generated a large group of mice expressing various combinations of wild-type and mutant ITAMs in the TCR-CD3 complex. A low number of ITAMs resulted in lethal autoimmune disease due to defective negative selection. A high ITAM number correlated with greater T cell proliferation while maintaining central tolerance. In another recent study he has shown that IL-35 is a novel inhibitory cytokine, specifically produced by Treg cells, which contributes for their suppressive activity. Ectopic expression of IL-35 confers regulatory activity on naive T cells, whereas recombinant IL-35 suppresses T-cell proliferation. For more information, visit The Immunology Interest Group |
