Cellular proteins associated with HIV-1 and SIV virions.

A knowledge of cellular proteins physically associated with HIV-1 and SIV is important for whole virus vaccines and understanding the disease process. To identify such cellular proteins, purified virus was disrupted and proteins separated by reversed-phase high-pressure liquid chromatography (rp-HPLC) and/or SDS-PAGE and identified by N-terminal amino acid sequence analysis. Whole virus was isolated (by density gradient centrifugation) from H9 or CEM cells infected with either HIV-1MN and also from HuT 78 cells infected with SIVMne. To date, a total of 40 cellular proteins have been identified. Some cellular proteins such as actin and ubiquitin are found in all viral preparations, while others such as the Class I and Class II histocompatibility antigens are only in virus stocks prepared from lymphoid cells (H9 or HuT 78). Virus produced from CEM cells is devoid of histocompatibility antigens but contains substantial amounts of other proteins such as glyceraldehyde 3-phosphate dehydrogenase. Thus, the number and nature of copurifying cellular proteins is dependent upon the cellular origin of the virus. Moreover, some proteins may be specific for the virus type. SIV preparations contain substantial amounts of Histone 3.1 and Histone 2B; whereas, HIV-1 contains only Histone 2B. Thymosin 4 is found with HIV-1 but not with SIV; however, both preparations contain Cyclophilin A. We are using 1) immune precipitation of intact virions, 2) virus neutralization, and 3) protease susceptibility, before and after disruption of the viral membrane, to determine if specific cellula proteins are physically associated with virions. These results and a list of viral associated cellular proteins will be presented.
