For taxonomic analysis of phytoplankton samples obtained, a 300-500 ml subsample of each collection was fixed with Lugol's soulution and concentrated by allowing the cells to settle for 72 hours. Only 100 ml subsample was used for 5 samples due to unusually high amounts of suspended particulate in these samples. After decanting the supernatant water, a concentrated subsample of 50 ml was transferred to an acrylic plastic chamber, and the contents were allowed to settle for 24 hours. The chamber then was mounted on an inverted microscope (a Wild M40 was used for the 1989-90 samples and a Nikon Diaphot-TMD was used from 1990-93) and 300 algal cells were identified and counted with 100x phase oil objective. Data obtained from the counting procedure were used to estimate relative adundances, cell densities, and cell biovolumes of taxa in water samples. Estimates of cell densities were obtained from measurements of the counting chamber, field lengths and widths of the observed transects, and corresponding dilution factors: algal units/liter = count x {A/(W x L)] / m[V/1000 ml] x CF (where A is the area of the chamber (cm squared), W is the field width (cm), L is the total length of the transects (cm), V is the volume of the chamber (ml) times 1 liter, and CF is the volume of the concentrated sample divided by the volume of the original sample). Biovolume conversion factors were determined for each taxon using appropriate geometric formulae. Cell densities were multiplied by these factors to obtain estimates of cell biovolumes.
