Technical Abstract: MicroRNAs are small ~22 nucleotides-long non-coding RNAs capable of controlling gene expression by inhibiting translation or targeting messenger RNA for degradation. Bovine genome sequence is not yet annotated for the microRNAs and there are currently no bovine miRNAs reported in the miRBase. Alignment of human microRNA sequences against the bovine genome resulted in the identification of 298 bovine miRs. In order to validate these predictions and identify new bovine microRNAs we have constructed and sequenced smallRNA cDNA libraries from bovine embryo, thymus, small intestine, abomasum, and mesenteric lymph nodes. This strategy allowed the identification of 130 bovine microRNAs. Of these, 101 aligned to known human microRNAs, 7 of them only aligned to the complementary strand of the mature microRNA in the database. The remaining 22 had secondary structures characteristic of microRNAs and 11 of these have phylogenetic conservation with other species.
