Research Project: Proteinase Activity and Malting Quality in Harrington X H. Spontaneum Recombinant Chromosome Substitution Lines

Objective:
Assay proteinase activity and characterize proteinase composition on Recombinant Chromosome Substitution Lines from malting barley x wild barley (Harrington x H. spontaneum) cross; use data to link proteinase phenotype to genotype, generating QTL or SNP genotype for protein mobilization component of malting quality.

Approach:
We propose to examine both serine proteinase activity and composition in a population (Recombinant chromosome substitution lines) developed from a cross of wild barley (H. spontaneum) with an elite malting barley variety (Harrington) to establish the progenitor proteinase activity (H. spontaneum) and identify those activities that appear to have been lost during development of the malting line. Proteinase activity and composition data will be leveraged with existing QTL and SNP genotyping of this population currently underway by collaborator.
