Tissue Preparation Overview

Antibody Purified

A specific population of cells is purified using an antibody, normally attached to a magnetic bead

Bulk Tissue

Whole tissue specimens (>0.5 g) which contain a variety of malignant and/or normal cell types. Tumor samples are selected to be enriched in tumor cells.

Cell Lines

Cell lines are cultured cells grown in vitro.

Flow Sorted

Flow sorted cells are cells separated by flow cytometry.

Micro-Bulk Tissue

Whole tissue specimens (<0.5 g) which are of biological interest, but cannot be utilized for traditional libraries due to their small size. Libraries are prepared by a PCR-based method employing 10 cycles of PCR. See Krizman Protocol 1.

Microdissected Tissue

An important goal of CGAP is to develop and evaluate methods to recover and analyze mRNA from microdissected tissue samples. This is accomplished by comparing and contrasting cDNA libraries using several different methodologies. The three methods that have been used to date are Krizman Protocol 1, Krizman Protocol 2, and Krizman Protocol R/D. Short-term Culture

Short-term cultured cells are cells grown in vitro, but a permenant cell line is not established. Thus, the cells might be more like the original than an established culture.
