Technical Abstract: Ninety-six plants from each of three alfalfa accessions were analyzed to compare the relative efficacy of molecular marker types for germplasm characterization. The three accessions: 'Aragon', 'Hunter Rivers', and 'Yonca', originated in Turkey, Australia, and Spain, respectively. Markers produced from amplified fragment length polymorphism (AFLP), random amplified DNA (RAPD), and amplified fragments of hypervariable regions of the chloroplast genome were compared. Amplification of hypervariable chloroplast DNA produced a higher frequency of low molecular wight markers within the Spanish accessions Yonca, distinguishing it from the other more similar accessions. Continuing research will determine the utility of these marker types for analyzing genetic diversity through cycles of germplasm conservation.
