Quantification of P24+ lymphocytes as a function of CD4 count.

OBJECTIVE: To quantify HIV-infected lymphocytes in individuals at different stages of infection. METHODS: Individuals with HIV disease (N=33) receiving antiretroviral therapy were grouped according to CD4 count: Group I (N=18), CD4 counts between 200 and 500/mm(3); and Group II (N=15), CD4 less than 200/mm(3). Addition of a phycoerythrin-labelled, CD4-specific, monoclonal antibody to the standard lymphocyte p24-FCA assay (J. Inf. Dis. 162:1018-1024, 1990), a flow cytometric measure of HIV infection, allowed quantification of lymphocytes with cell-associated HIV-p24 antigen (P24+ lymphocytes) in both the CD4+ and CD4- lymphocyte compartments. TABULAR DATA, SEE ABSTRACT VOLUME. DISCUSSION AND CONCLUSION: Lymphocyte p24-FCA can be used to monitor HIV infection of lymphocyte subsets at different stages of infection. As CD4 counts fall, the percentage of p24+ cells rises, indicating an increasing viral load.
