Research Project: LOCALIZATION OF GENE EXPRESSION ASSOCIATED WITH TUBER PERIDERM MATURATION AND QUALITY

Objective:
The objective is to determine the cellular expression/localization profile of genes that are marker candidates for potato postharvest quality features. These markers include those involved in periderm maturation and skin-set in native and wound periderm.

Approach:
This cooperative research project involves follow-on experiments and assessments from a prior SCA. Nucleic acid-based techniques will be applied to determine localization of expressions of candidate genes involved in tuber skin-set and related postharvest quality features. In vitro grown minitubers and/or certified seed, nuclear or G1 generation seed tubers, will be used to investigate localization of gene expression via in situ hybridization and related techniques. Tissue and cell specific expression will be determined via one or a combination of dark field, bright field or fluorescence microscopy. Expression patterns will be localized during maturation of native and wound periderm. Results will help confirm involvement of the selected genes in periderm maturation and skin-set.
