Préparation de la prochaine génération de scientifiques
Pendant 30 ans, les bourses conférées par le NIH pour la recherche aux États-Unis ont permis aux universités et aux centres de recherche de préparer et de former la prochaine génération de scientifiques pour travailler dans les domaines de la recherche biomédicale, comportementale et clinique. La rubrique NIEHS News (p. A348) examine quelques unes des réussites issues de ces programmes.
La sécurité chimique aujourd’hui
Cette année marque le 20ième anniversaire de l’incident de l’usine de pesticides d’Union Carbide à Bhopal, Inde, largement considéré comme étant le pire accident industriel de l’histoire. Des mesures ont été prises pour assurer qu’un accident comme celui qu’a connu Bhopal ne se reproduise plus jamais, mais est-ce que cela suffit? La rubrique Focus (p. A352) examine la question de savoir si l’industrie chimique est réellement moins dangereuse qu’il y a 20 ans.
Rachat de terrains pour protéger l’environnement: une bonne ou une mauvaise affaire?
Ce mois-ci, la rubrique Sphères d’influence (p. A360) examine le pour et le contre des rachats de terrain. Ces rachats paraissent simples à première vue : l’achat d’un terrain permet de le protéger contre des activités destructrices pour l’environnement et de le maintenir propre et à l’abris de la pollution. L’expérience a néanmoins montré que la réalité n’est pas toujours aussi claire et nette.
La caméra cachée expose l’exposition
La rubrique Innovations (p. A364) examine le contrôle, par vidéo, des expositions en laboratoire. Cette technologie combine des images vidéo et des détecteurs de données chimiques, biologiques, radiologiques et physiques en temps réel pour mieux comprendre comment et quand les expositions dangereuses se produisent en laboratoire.
Recherche
Stockage de PCB dans des récipients en plastique et en verre
Karmaus et Riebow (p. 643) ont décelé une source potentielle de biais dans la détermination des concentrations d’organochlorures sériques. Le stockage du sérum dans des récipients en plastique a modifié les concentrations totales des polychlorobiphényles (PCB), notamment des PCB plus riches en chlorure, comparé à des échantillons stockés dans des récipients en verre. Les auteurs conseillent d’utiliser les méthodes standardisées pour le prélèvement, la manipulation, le stockage et la mesure des échantillons de nouveaux produits.
Inhibition des systèmes de signalisation par les perturbateurs endocriniens
La communication cellulaire conservée par le processus de l’évolution à l’intérieur d’un organisme et entre organismes dépend de l’interprétation efficace et fidèle des signaux chimiques par les récepteurs. Fox (p. 648) étudie des questions d’évolution courantes relatives à la signalisation chimique, telles que la signalisation des oestrogènes chez les vertébrés et la signalisation des phyto-oestrogènes des plantes aux bactéries, et suggère que de nombreux systèmes de signalisation constituent des cibles pour les perturbateurs endocriniens.
Une rétrospection de l’évolution des PBDE
Sjödin et al. (p. 654) ont mesuré six diphényléthers polybromés (PBDE), un hexabromobiphényle et un hexachlorobiphényle dans 40 pools de sérum humain prélevé dans le sud-ouest des États-Unis entre 1985-2002 et à Seattle, Washington, entre 1999-2002. Les concentrations de la plupart des PBDE augmentent. Les biphényles polychlorés et polybromés étaient en corrélation négative avec l’année de prélèvement, ce qui indique que les taux baissaient depuis leur élimination progressive dans les années 70.
Interaction synergique dans les expositions microbiennes
Nous en savons peu sur les interactions inflammatoires entre les différents microbes et leurs métabolites. Huttunen et al. (p. 659) ont étudié les réactions inflammatoires des macrophages de souris après une exposition aux six microbes de l’air ambiant seuls ainsi qu’aux six microbes ensemble avec l’actinomycète Streptomyces californicus. Les expositions combinées à quelques microbes et aux toxines avec le Str. californicus ont provoqué des réactions inflammatoires synergiques dans des taux de certaines cytokines proinflammatoires.
Dégradation oxydative de l’ADN chez des travailleurs exposés au PM2.5
L’huile résiduelle riche en métal de transition (ROFA) est un mélange complexe comprenant des métaux qui peuvent être carcinogènes. Kim et al. (p. 666) ont étudié l’association entre l’exposition particulaire 2,5 µm (PM2.5) et la dégradation oxydative de l’ADN révélée par les concentrations urinaires de 8-hydroxy-2´-désoxyguanosine (8-OHdG) chez des chaudronniers exposés au ROFA et à des vapeurs comprenant des polluants métalliques. Les taux urinaires de 8-OHdG étaient sensiblement plus élevés dans les échantillons prélevés après la période de travail comparés aux échantillons prélevés avant la période de travail, après les ajustements en fonction des taux urinaires de cotinine, de l’état de bronchite chronique et de l’âge.
Inhibition de la signalisation symbiotique par les perturbateurs endocriniens
Certains perturbateurs endocriniens synthétiques imitent les hormones dans un grand nombre de classes de vertébrés. Une signalisation équivalente à celle des hormones est également observée lorsque les vertébrés sont exposés à des substances végétales appellées phyto-oestrogènes. Fox et al. (p. 672) montrent que certains pesticides et des perturbateurs endocriniens organochlorés connus pour la perturbation de la signalisation endocrinienne de la faune et la flore et chez les humains, perturbent également la signalisation des phyto-oestrogènes utilisée par les légumineuses pour attirer des bactéries du sol pour la fixation symbiotique de l’azote. Les principes sous-jacents de la perturbation endocrinienne pourraient être bien plus importants du point de vue écologique que ce que l’on avait pensé. (Voir également Sélections scientifiques, p. Le métabolisme du méthyleugénol chez l’Homme
Le méthyleugénol est structurellement semblable au safrol et est un carcinogène connu chez les rongeurs. Schecter et al. (p. 678) ont étudié le métabolisme humain à l’aide de neuf volontaires hommes et femmes qui ont reçu des biscuits au gingembre commerciaux dont la concentration de méthyleugénol était élevée. Le taux moyen de méthyleugénol dans le sérum à jeun était de 16,2 ± 4,0 pg/g de poids humide. Le taux sanguin avait atteint son maximum (53,9 ± 7,3 pg/g de poids humide, moyenne ± SD) après 15 min, suivi par un déclin rapide. La demi-vie d’élimination était d’environ 90 min.
Contrôle de composés perfluorés chez des mulots
Des composés perfluorés ont récemment été observés chez diverses espèces de la flore et la faune aquatique, des mammifères piscivores et des oiseaux. Hoff et al. (p. 681) ont étudié des mulots (Apodemus sylvaticus) capturés dans les environs immédiats d’une usine de composés fluorés (Blokkersdijk, Belgique) et d’un endroit plus éloigné. Les concentrations d’acide de perfluorooctane sulfonate (PFOS) dans le foie chez les mulots de Blokkersdijk étaient extrêmement élevées (0,47-178,55 µg/g de poids humide). Les résultats semblent indiquer une indépendance sexuelle des taux hépatiques de PFOS, des taux élevés d’accumulation de PFOS chez des mulots plus âgés et le transfert de PFOS in utero aux petits.
Champs magnétiques et cassures de brins de l’ADN
Des rats intensément exposés à un champ magnétique sinusoïdal de 60-Hz à des intensités de 0,1-0,5 mT permettent de mettre en évidence des cassures de simple et double brin de l’ADN dans les cellules cérébrales. Lai et Singh (p. 687) constatent que l’effet est cumulatif lors d’une exposition prolongée. Le Trolox (analogue de la vitamine E), le 7-nitroindazole (inhibiteur de l’oxyde nitrique synthase) ainsi que la défériprone (chélateur du fer) ont tous bloqué la cassure des brins de l’ADN provoquée par le champ magnétique. Les auteurs envisagent que les processus à médiation de fer, tels que la réaction de Fenton, intensifient la formation de radicaux libres, conduisant à des cassures de brins de l’ADN et à la mort cellulaire. Comparaison inter-laboratoires de recherches de l’androgénie
Dans le cadre d’une étude inter-laboratroires internationale, Körner et al. (p. 695) ont évalué et comparé quatre dosages in vitro à l’aide de deux androgènes stéroïdiens, de deux anti-androgènes, d’un témoin androgène (5-dihydrotestostérone) et d’un témoin anti-androgène (bicalutamide, ou ICI 176,334) pour déceler des agonistes et des antagonistes androgènes. En résumé, tous les dosages se sont avérés être des outils de dépistage utiles pour la détection et la quantification des effets androgènes et anti-androgènes médiés par AR de ces substances chimiques.
Traumatismes in utero des embryons exposés à des pesticides
Greenlee et al. (p. 703) ont évalué des expositions à des doses faibles de produits chimiques à usage agricole courant et à des pesticides utilisés pour l’entretien du gazon pour connaître les effets directs sur des embryons de pré-implantation de souris in vitro, ce qui correspond à 5-7 jours de post-conception chez l’Homme. Les résultats témoignent que le traumatisme provoqué par un certain nombre d’agents contenus dans les pesticides, à des concentrations supposées ne pas avoir des effets défavorables sur la santé humaine, peut se présenter très tôt in utero.
Extrapolation rétrograde et évaluation de l’exposition
Les être humains sont exposés à des taux de contamination continue d’organochlorés par la consommation de poissons pêchés pour le sport dans les Grands Lacs. Karmaus et al. (p. 710) ont comparé trois modèles d’extrapolation rétrograde pour évaluer les inconvénients possibles provoqués par des expositions antérieures à l’aide de mesures recueillies dans les années 70, 80 et 90. Les résultats suggèrent le besoin d’agir avec prudence lors de l’utilisation de techniques simples d’extrapolation rétrograde pour évaluer l’exposition dans des situations dont l’exposition aux polluants peut changer. Chaque fois que des mesures en série sont disponibles, les analyses de régression semblent produire des estimations rétrogrades plus précises de l’exposition.
Le stress maternel modifie les effets in utero du plomb
Les effets d’un statut socioéconomique défavorable utilisés comme facteur de risque pour la maladie et les dysfonctionnements ont été attribués au stress chronique et à une élévation de glucocorticoïdes. Une élévation chronique de glucocorticoïdes et de plomb provoque des modifications de comportement analogues. Cory-Schlecta et al. (p. 717) se sont servis d’un modèle rongeurs pour étudier les contributions du stress maternel (contrainte) et de l’exposition maternelle au plomb aux taux de corticostérone des petits, aux modifications des neurotransmetteurs et à une ligne de base du comportement. La potentialisation des effets du plomb et du stress soulèvent des questions au sujet des stratégies actuelles pour réduire les risques.
Conséquences de la catastrophe du WTC
Landrigan et al. (p. 731) caractérisent les expositions et les inconvénients appréciables découlant de la destruction du World Trade Center (WTC). Des femmes enceintes se trouvant à l’intérieur ou près du WTC le 11 septembre 2001 ont montré deux fois plus de nouveau-nés qui présentaient un retard de croissance intra-utérin. La poussière du WTC a provoqué une hyperréactivité bronchique, une toux persistante et un risque accru d’asthme. Le risque futur de mésothéliome pourrait être élevé, notamment parmi les personnes exposées à l’amiante. Il est recommandé de continuer le suivi de toutes les populations exposées.
La génétique humaine et la justice environnementale
Sze et Prakash (p. 740) résument la conférence organisée à New York sur les implications éthiques et sociales de la génétique humaine, de l’environnement et des communautés de couleur ainsi que la réunion de travail sur la génétique humaine et la justice environnementale. L’objectif de la conférence était de sensibiliser les participants à la science et à l’éthique de la recherche génétique dans le contexte de l’hygiène du milieu et des communautés de couleur. Le colloque proposait un débat sur les idées et les préoccupations principales des défenseurs communautaires de la justice environnementale quant à la recherche génétique et l’hygiène du milieu.
Médecine environnementale
Hémochromatose et plomb
Le taux fer dans le corps est inversement associé à l’absorption du plomb, et les gènes associés à l’hémochromatose peuvent modifier le taux de plomb dans le corps. Wright et al. (p. 746) ont effectué une étude pour vérifier si les gènes de l’hémochromatose (HFE) C282Y et/ou H63D étaient associés aux taux de plomb de la rotule et du tibia chez des sujets d’une étude normative sur le vieillissement. Dans les analyses multifactorielles, suite aux ajustements en fonction de l’âge, le tabagisme et l’éducation, la présence d’une variante d’allèle de HFE était un indice indépendant de taux de plomb de la rotule nettement plus bas (p < 0,05).
Médicaments et phtalates
Les phtalates sont utilisés dans des produits de consommation et de soins personnels ainsi que dans les produits en plastique et les dispositifs médicaux, cependant les informations sur les sources d’exposition des êtres humains aux phtalates sont limitées. Hauser et al. (p. 751) citent le cas d’un malade ayant reçu de l’Asacol pour traiter une rectocolite hémorragique. Un échantillon urinaire ponctuel recueilli 3 mois après le début du traitement a révélé que la concentration de mono-butyl phtalate du malade était de 16 868 ng/ml—soit, plus de deux ordres de grandeur supérieures au 95ième percentile signalé pour les hommes dans l’enquête sur la santé et le régime aux États-Unis (National Health and Nutrition Examination Survey) de 1999-2000.
Expositions personnelles au PAH
Tonne et al. (p. 754) ont mesuré les expositions personnelles à neuf hydrocarbures aromatiques polycycliques (PAH) parmi 348 Afro-Américaines et Dominicaines non-fumeuses enceintes pendant leur troisième trimestre de grossesse. Les concentrations moyennes de PAH variaient entre 0,06 ng/m3 pour le dibenz[a,h]anthracène et 4,1 ng/m3 pour le pyrène, et la concentration moyenne du benzo[a]pyrène était de 0,50 ng/m3. Les concentrations de la plupart des PAH étaient plus élevées en hiver qu’en été. Une analyse de régression linéaire multiple a mis en évidence des associations entre les expositions au PAH et le temps passé dehors, le chauffage d’habitation et de l’encens allumé à l’intérieur.
Expositions tôt dans la vie et asthme chez l’enfant
Salam et al. (p. 760) ont réalisé une étude cas-contrôle sur l’asthme dont les cas était définis comme souffrant de l’asthme (diagnostiqué par un médecin) dès l’âge de 5 ans et les contrôles ne souffrant pas d’asthme lors de l’inclusion dans l’étude. Les contrôles étaient souvent appariés suivant l’âge, le sexe et la communauté de résidence, et opposés par rapport à l’exposition in utero au tabagisme de la mère. L’asthme diagnostiqué chez des enfants avant l’âge de 5 ans était associé à des expositions lors de la première année de vie à la fumée de bois ou de mazout, à la suie ou aux gaz d’échappement, à des cafards, à des herbicides, à des pesticides, ainsi qu’à des produits agricoles, de la poussière, ou des animaux d’une ferme.
Leucémie chez l’enfant et installations de forces aériennes
Une série exceptionnelle de cas de leucémie chez l’enfant s’est produite près de la ville de Fallon dans le comté de Churchill, Nevada, entre 1999 et 2001. Steinmaus et al. (p. 766) ont fait des recherches pour savoir si la série était le fait du hasard ou si elle était éventuellement liée à des expositions provenant d’une base aéronavale dans les environs. Les auteurs ont également comparé des taux de leucémie chez l’enfant d’autres comtés aux États-Unis ayant des installations de forces aériennes. Les résultats attestent que la série du comté de Churchill n’était probablement pas le fait du hasard, cependant aucune augmentation générale de leucémie chez l’enfant n’a été trouvée dans d’autres comtés des États-Unis ayant des bases d’aviation militaires.
