1H NMR studies on HIV interaction with T and NK lymphocyte subpopulations.

INTRODUCTION: In previous work, we have demonstrated that the changes in 1H NMR spectra occurring during HIV infection of susceptible cells reveal membrane structure modifications during viral internalization, thus providing a clear picture of the early events of infection. Aim of this study is the analysis of HIV interaction with different lymphocyte subpopulations by means of NMR spectroscopy, in other to provide more insight on cell tropism. METHODS: The changes in NMR signals arising from mobile lipid domains, used as natural probes for membrane structure, were quantitated. Activated lymphocyte subpopulations T4, T8 and LAK cells were separated by means of immunobead adsorption. Analysis for surface markers was performed by flow cytometry. Contamination was less than 4%. Samples of 15 x 10(6) cells were infected at 0.01 TCID50/cell with HIV-1, HTLV IIIB strain. RESULTS: Changes in the intensity of the fatty acid peaks in NMR spectra were detected accompanying HIV entry into the membrane of PHA stimulated lymphocytes and compared to the effects observed in the susceptible T4 subset. Interaction of the virus with homogeneous populations of T8 cells failed to induce any effect on the fatty acid signals, thus confirming that resistance to HIV infection for T8 cells is due to failure of viral particles to penetrate into the cell membrane. On the contrary, NMR revealed very deep perturbations in the lipid signals from the membrane of LAK cells after incubation of 90 degrees with HIV, thus indicating that the viral particles enter these cells irrespective of the presence of the receptor.
