Entrez Gene Number:
Entrez Gene Summary
PPP1R14A is a phosphorylation-dependent inhibitor of smooth muscle myosin phosphatase (see MIM 603768). Inhibition leads to increased myosin phosphorylation and enhances smooth muscle contraction in the absence of increased intracellular Ca(2+) concentration.[supplied by OMIM]

Descriptive Text:
The myosin phosphatase inhibitor protein, CPI-17, is expressed in smooth muscle, platelets and brain, and its phosphorylation at Thr38 enhances inhibitory potency 1000-fold. CPI-17 is an established substrate for PKC, ROCK, PKN, and ZIP-like kinase. It plays a central role in control of myosin phosphatase. Platelet activation and smooth muscle contraction in response to histamine both trigger phosphorylation of Thr38. Addition of CPI-17 to skinned smooth muscle restores PKC-stimulated contraction and expression of CPI-17 in fibroblasts is thought to cause stress fiber bundling.
