Approximately 50% of breast and ovarian cancers can be accounted for by mutations in the breast and ovarian cancer susceptibility genes BRCA1 and BRCA2. BRCA1 encodes a nuclear phosphoprotein which functions as a tumor suppressor. Transfection of BRCA1 into transformed cells has been reported to flatten morphology and reduce anchorage-independent growth, while expression of an anti-sense construct in normal fibroblasts achieves the reverse. BRCA1 works closely with Rad51, which plays an important role in repairing genetic damage in cells, suggesting a role for BRCA1 in the control of DNA recombination and of genome integrity.
