Experimental Design A PolII-neomycin resistance cassette which lacked a polyadenylation signal was inserted into exon 5 of p53. The insertion was accompanied by a small deletion of a 450 bp fragment containing 106 nucleotides of exon 5 and 350 nucleotides of intron 4.  
Phenotype Mice homozygous for the mutated p53 gene are viable and appear completely normal but are susceptible to the spontaneous development of different types of tumors before they reach 6 month of age, particularly lymphomas and sarcomas. Heterozygote progeny carrying single wild-type p53 allele rarely develop spontaneous tumors before they are 9 month old.
