Description: |
A GFP reporter cDNA was inserted by homologous recombination into the Arf-specific exon 1 beta of the Cdkn2a locus. Expression of p19ARF is disrupted and GFP is expressed under control of the endogenous Arf promoter. Homozygous Arf animals spontaneously developed tumors by 16 weeks of age. The predominant tumor types were highly invasive sarcomas of various types (65%) and lymphomas (33%), similar to Arf-null mice (see MMHCC strains 01XF6 and 01XG7). Depending on tumor type, size, and anatomical location, many solid tumors exhibited vivid green fluorescence, and metastatic lymphomas sometimes appeared macroscopically as green fluorescent nodules in the liver, indicating that the Arf promoter remained active in late stages of tumor development. The ArfGFP mutation allows for tumor progression to be monitored by whole-body fluorescence imaging. |
