Strain Description: The mdm2puro allele is a hypomorphic and conditional allele that can be converted to either a fully functional conditional allele or a null allele by the Cre recombinase. Mice expressing mdm2puro (Tac125) allele and one known null allele of mdm2 have reduced levels of expression of the oncogene mdm2 and, as a consequence, are small, radiosensitive and lymphopenic. All phenotypes are dependent on p53. Although the transcriptional activation function of p53 is increased in many tissues, spontaneous, p53-dependent apoptosis occurs in only a subset of these tissues. These mice will be valuable for identifying the regulatory steps that control p53 function in tissues.
