Description: |
Doxycycline-induced knockouts of a floxed allele can be engineered in mice containing Fabpl4X at -132/rtTA and TetO-Cre. Analysis of recombination using the floxed R26R Cre reporter indicates that the principal site of recombination is in the proximal two thirds of the small intestine with lesser degrees of recombination distally. Recombination occurs in long-lived multipotent stem cells and all descendant lineages. Note that recombination is mosaic: some crypts support recombination while others do not (see Wong et al., PNAS 97:12601 (2000) for illustration. There is no expression in the urothelium or in other extraintestinal sites except the liver: a subset of tri-transgenic mice hemizygous for Fabpl4X at -132rtTA, tetO-Cre and R26R exhibit recombination of R26R in scattered groups of hepatocytes after doxycycline treatment. No recombination is observed in any organ surveyed in the absence of doxycycline. |
