Abstract: Enzyme-catalyzed O-18 labeling appears well-suited for sensitive quantitative comparisons. (The global introduction of isotope labels at the peptide stage mandates that protein manipulation be minimized, so as to minimize differential losses. Fractionation should be optimized for the combined pools of peptides labeled with light and heavy isotopes.) In the present study, forward and reverse O-18 labeling are integrated with solution isoelectric focusing and capillary LC-tandem mass spectrometry to study changes in mitochondrial proteins associated with drug resistance in human cancer cells. The reverse labeling experiment was carried out to provide complementary pairs of isotopically labeled peptides. These were used to estimate the precision and dynamic range of the method, and to confirm the behavior of on/off proteins.
