Research Interests
Visualization of biological structure at the nanoscale. In particular:
- Electron tomography - reconstruction from multiple views of the sample in order to visualize subcellular structure in 3D. Tomography of stained plastic sections and of frozen-hydrated material, with the eventual goal of observing macromolecular complexes inside hydrated cells and tissues.
- Localizing macromolecules and chemical elements, using electron microscopy, within the context of an intact cell. Immunogold labeling, cytochemistry, in situ hybridization, X-ray microanalysis, and EELS.
- Advances in methods of specimen preparation, especially those facilitating observation of biological material in a frozen-hydrated, life-like state.
- Computer image processing and analysis. 3-D reconstruction and feature extraction algorithms. Scripting to streamline and automate routine tasks.
