2520: Comprehensive Assessment of Long-term Effects of Reducing Intake of Energy (CALERIE)

Researchers will explore whether healthy adults who eat a 25% calorie restricted diet for two years will experience biological changes thought to slow the aging process and protect against age-related disease.

Healthy Men ages 20-50, and Women ages 20-47

The effects of calorie restriction have been studied in animals for years, and this is the only intervention shown to slow the aging process, extend lifespan, and maintain health and vitality. The specific health benefits could include such things as lowering the risk of developing diabetes or cardiovascular disease. It is not known if people will have the same benefits as seen in animals. This is what CALERIE will examine.
Major Exclusions:

• BMI not within 22-27.9 kg/m2
• Smoking
• Diabetes
• Cardiovascular disease
• Plans to move from Boston during the study period
• Pregnant or breast feeding, if female or planning to become pregnant during the study period.

Length of Study: 24 months. # of Visits: Assessment visits are scheduled at Baseline, 1, 3, 6, 9, 18, and 24 months of participation.
Residency: Total of 8 overnight stays Study diet

Volunteers randomized into the calorie restricted group will reduce calorie intake by 25% of baseline which is expected to cause weight loss, though this is not guaranteed.

The first month of food is provided by the research center. Thereafter, group and individual meetings with nutrition and behavioral coaches will assist volunteers with implementing the eating plan to best fit their lifestyle.

Volunteers randomized into the non-calorie restricted group will maintain regular eating habits.

Tests:
  • Energy Metabolism
  • Body composition and bone health
  • Cardiovascular fitness, muscle strength and endurance
  • Cognitive functioning
  • Quality of life questionnaires
  • Cardiovascular and diabetic risk assessments
