Objective:
Determine placental involvement in transmission of experimental BSE in sheep.
Approach:
Orally dose a group of susceptible (QQ) ewe lambs with inoculum from the brains of cattle infected with BSE. Breed the ewes and collect the placentas from the lambings. The placentas will be tested for PrP-d by Western blot and IHC. VLA will do the oral dosing, collect the samples, and perform the Western Blots. VLA will send the formic acid treated samples in paraffin blocks to Pullman for IHC. ARS will perform the IHC staining with appropriate primary antibodies for PrPd detection.
