Trial Description

Purpose:

Drugs used in chemotherapy, such as capecitabine, fluorouracil, and oxaliplatin work in different ways to stop tumor cells from dividing so they stop growing or die. Radiation therapy uses high-energy x-rays to damage tumor cells.

This randomized phase III trial is studying radiation therapy and either capecitabine or fluorouracil with or without oxaliplatin and comparing them to see how well they work when given before surgery in treating patients with resectable rectal cancer. It is not yet known whether radiation therapy and either capecitabine or fluorouracil is more effective with or without oxaliplatin in treating rectal cancer.

Eligibility:

Eligibility criteria include the following:

• No metastatic disease
• No previous radiation therapy to the pelvis

Final eligibility for a clinical trial is determined by the health professionals conducting the trial.

Treatment/Intervention:

Patients will be randomly assigned (have an equal chance of being placed) to one of four groups.

Patients in group one will receive a 5-day continuous infusion of fluorouracil and undergo radiation therapy 5 days a week for 5-6 weeks.

Patients in group two will receive a 5-day continuous infusion of fluorouracil and undergo radiation therapy 5 days a week for 5-6 weeks. They will also receive a 1-hour infusion of oxaliplatin once a week for 5 weeks.

Patients in group three will receive capecitabine by mouth twice a day 5 days a week and undergo radiation therapy 5 days a week for 5-6 weeks.

Patients in group four will receive capecitabine by mouth twice a day 5 days a week and undergo radiation therapy 5 days a week for 5-6 weeks. They will also receive a 1-hour infusion of oxaliplatin once a week for 5 weeks.

Within 6-8 weeks after the completion of chemotherapy and radiation therapy, all patients will undergo surgery. Quality of life will be assessed periodically.

After completion of study treatment, patients will be evaluated every 6 months for 5 years.
