Abstract: We tested the hypothesis that early polio vaccines contaminated with Simian virus 40 (SV40) increased the risk of brain tumors using data from a large case-control study of adult brain tumors conducted in the US between 1994 and 1998. Risks for both injected and oral vaccine given during the 1954-1962 period, when they were most likely contaminated, and for birth years 1941-1962, when the majority were exposed to the potentially contaminated vaccine during infancy or early childhood, were around unity. This is consistent with previous epidemiological studies, however future studies should incorporate new genetic and serological markers of individual SV40 exposure to clarify its role in very rear types of brain tumors.
