Research Project: TAILORING SUPERIOR ALLELES OF ALTSB, A MAJOR SORGHUM ALUMINUM TOLERANCE GENE, FOR DEPLOYMENT INTO CROP BREEDING PROGRAMS

Objective:
1) Survey AltSB alleles across diverse sorghum germplasm; 2) Identify superior AltSB haplotypes by association mapping; 3) Develop and use haplotype-specific markers for introgression into pre-breeding near-isogenic lines; and, 4) Survey sorghum homologs of novel candidate Al tolerance genes we have identified from complementary work in maize, wheat and Arabidopsis. 425 log 32530 approved December 2006.

Approach:
In the Kochian lab, we will work on the association mapping of the region of the sorghum genome where the sorghum Al tolerance gene, AltSB, is located, to identify polymorphisms associated with enhanced gene expression and protein function resulting in stronger Al tolerance. This involves sequence this region in a panel of 240 diverse sorghum lines, and phenotyping the same lines for Al tolerance, AltSB expression, and Al-activated root citrate exudation.
