Objective:
To characterize the genetics of host-pathogen interactions in both fungal and viral pathogen systems in order to develop a better understanding of resistance and pathogenicity. Previous work to better characterize these interactions will be extended and expanded to include identification of candidate genes associated with avirulence/virulence loci in segregating Pyrenophora teres pathogen populations.
Approach:
A population consisting of P. teres parents displaying different virulence patterns on various barley lines will be used to clone avirulence genes involved in net blotch disease. A moderately saturated mapping population of 78 progeny has been used to identify 3 avirulence loci in a single population. Phenotypic data along with molecular marker data will be generated in another 300 progeny in order to develop a more saturated map suitable for map-based cloning. BAC libraries will be developed for each parent. Once closely-linked flanking markers are identified for each avirulence gene, BAC clones will be identified and end sequencing will be done to begin development of a contig for identification of candidate genes.
