Technical Abstract: The polymorphic fungus Aureobasidium pullulans was isolated from many provinces in tropical Thailand and cultured in pullulan production medium (PM) containing sucrose and peptone as carbon and nitrogen sources, respectively. Liquid PM cultures varied in color from cream to pale pink, light burgundy red, light brown, olivaceous green, and black. Most isolates produced colonies that were smooth, moist, and cream or pale pink on yeast malt agar (YMA) medium. Molecular sequence analyses of the internal transcribed spacer (ITS) regions of rDNA showed no differences among these isolates. However, the intergenic spacer (IGS) regions of rDNA revealed the possibility of strain differences. Preliminary data from 9 isolates suggest that IGS-PCR patterns cluster in groups according to color variability. Sequence analysis of 3-4 Kb of IGS1 and IGS2 showed similarities among isolates consistent with the IGS-PCR patterns.
