EPA Grant Number: R831711C003
Subproject: this is subproject number 003 , established and managed by the Center Director under grant R831711
(EPA does not fund or establish subprojects; EPA awards and manages the overall grant for this center).

Center: Mount Sinai Center for Children’s Health and the Environment
Center Director: Wolff, Mary S.
Title: Genetics of Phthalate and Bisphenol A Risk in Minority Populations (Individual Susceptibility)
Investigators: Wetmur, James G.
Institution: Mount Sinai School of Medicine
EPA Project Officer: Fields, Nigel
Project Period: November 1, 2003 through October 31, 2008
Project Period Covered by this Report: November 1, 2005 through August 23,2006
RFA: Centers for Children's Environmental Health and Disease Prevention Research (2003)
Research Category: Health Effects , Children's Health

Description:

Objective:

The objective of this research project is to study seven genetic polymorphisms in the enzymes that activate and detoxify organophosphates and other pesticides in the population of mothers and infants enrolled in project R831711C002.

Progress Summary:

Project 3 has completed genotyping for a variant related to bisphenol A metabolism and analysis of those data is in progress. Genotypes and phenotypes of PON1 have been assessed in 656 samples, including maternal and cord bloods. Genotype/haplotype-phenotype associations of PON1 were found to vary by allele, to be independent of race/ethnicity, and to be stronger for infants than for mothers.

Future Activities:

We shall continue to collaborate with Project 2 to examine PON1 genotype and phenotype effects on developmental outcomes. We will complete the predictor paper on UGT2B7 genotype and maternal phenols in urine. We will also examine associations of the common UGT2B7 gene variant with birth outcomes and neurodevelopment with Project 2. The UGT2B7 promoters of our African-American population will be resequenced. The lipase/amylase data collection will continue.
