Objective:Over the past few decades, concerns regarding the levels of trace metals in water systems have risen because, unlike some organic pollutants, they present a recirculating environmental problem. For highly contaminated sites, the majority of metal remediation would need to be achieved through precipitation and filtration. However, these techniques cannot attain the low levels imposed by regulatory requirements. To comply with these legal limits, a "polishing" step involving chemical extraction is needed. While organic contaminants can be extracted and/or decomposed by thermal or biological means, metals must be eliminated by reclamation. Typically, these reclamation efforts focus on the use of ion exchangers. Ideal ion exchangers should exhibit the following characteristics: high selectivity for the target metal of interest, strong binding with a large capacity, fast binding kinetics, efficient, on-demand metal release, and minimal initial and operating costs. Due to the contradictory nature of some of the ideal attributes of ion exchangers, e.g. strong binding capabilities and easy release, many of the traditionally used synthetic organic exchangers fail to satisfy all of the model characteristics. In addition, many of these organic exchangers, such as crown ethers, require harsh, expensive treatments to reclaim the target metals. Chelex-100, a commonly used exchange resin, does demonstrate selectivity against alkali metals. However, it offers little selectivity amongst the transition metals. Additionally, many ion exchangers exhibit lower effective exchange capacity in salty matrices. To overcome the drawbacks of traditional ion exchangers, new technology is being explored using short chain biopolymers made up of single amino acids (i.e., n=50) immobilized on solid support systems (i.e., controlled pore glass, CPG). The polymer is chemically bonded to the surface producing a chelator capable of three degrees of freedom for metal binding. These polymers exhibit strong binding and on-demand release by a simple change in pH. They can also achieve selectivity by altering the amino acid side chain. Additionally, the solid support is not subject to the degree of shrinkage and swelling seen in many of the traditionally used exchange resins. The first two biopolymers explored have been poly-L-cysteine (PLCys), a soft acid metal binder, and poly-L-aspartate (PLAsp), a hard acid metal binder.
Summary/Accomplishments (Outputs/Outcomes):Preliminary research into the use of PLCys for metal remediation efforts began by characterization of homogeneous PLCys and the application of a flow injection system. Following initial research, a full characterization of the PLCys-CPG system was undertaken. As a comparison study, the performance of PLCys as a metal chelator was evaluated against a traditional exchanger, 8-hydroxyquinoline. The chemically reactive nature of PLCys and the role its thiol disulfide bonds play in metal binding were then explored more intensively. Following the complete characterization of the PLCys-CPG system, a new system, PLAsp, was investigated. PLAsp was then contrasted to the chelation capabilities of a similar, synthetic polymer - poly-acrylic acid. To better understand the mechanics of binding in these poly-amino acid systems, an in situ study was completed utilizing tapping mode atomic force microscopy to quantitate and visualize changes in the polymer in different environments. In an effort to maximize metal selectivity, attempts were made to template PLCys for target metals by using disulfide bonds. In order to broaden the possibilities for further applications, studies into alternative support systems, such as gold, were then completed. Finally, to further understand the nonequilibrium binding in these microcolumns, a model was developed to predict binding behavior of various metals.
