Research Project: IDENTIFICATION OF PROTECTIVE INNATE IMMUNE MECHANISMS IN COCCIDIOSIS AND SALMONELLOSIS

Objective:
To identify host genes which control innate immune responses to intracellular pathogens such as Eimeria using macrophage and intestinal cDNA microarray.

Approach:
Macrophage and intestinal cDNA arrays will be used host genes which are expressed during coccidiosis and salmonellosis. RNA samples from macrophages and intestinal lymphocytes infected with Eimeria (ARS-BARC) Salmonella (FDA) will be prepared and sent to the University of Delaware. At least 4 different time points using 3 different Eimeria and different wild and mutant types of Salmonella will be used in this study. Dr. Calvin Keeler will conduct avian macrophage cDNA microarray to identify host genes which control innate immunity mediated by macrophages in coccidiosis and salmonellosis (University of Delaware). Validation of microarray data and investigation of the role of host immune factors which were identified in the microarray studies will be carried out at BARC using RT-PCR.
