Technical Abstract: Numerous studies have established that recombinant bovine Somatotropin (rbST) will stimulate growth in the rainbow trout. However, the effects of rbST on target tissue gene expression are not well characterized. In the current study, we used PosilacĀ® (Monsanto Co., St. Louis, MO) to assess the effects of an rbST implant on liver and muscle transcript levels in fish selected for high growth rate. Twelve fish (~100 g bw) from each of two families selected for their high growth rates were placed into individual tanks. Following a two week acclimation period, four fish from each family received an injection of PosilacĀ® (120 micro g/g BW) and four received an equivalent volume of the carrier (~50 micro l). Three days after the injection, fish were euthanized and tissue and blood samples were collected. Blood IGF-I levels were used to verify increased growth hormone activity in the rbST treated fish. Alterations of muscle and liver transcriptomes by rbST were characterized using the MSU 1RT rainbow trout and GRASP 16K Atlantic salmon cDNA microarrays. Carrier injected samples were randomly paired with rbST injected samples within each family using a dye swap microarray design. Differentially-expressed transcripts will be explored for their utility as candidate genes associated with growth and development in the rainbow trout.
