Objective:
Develop a robust, high-throughput Sheath Blight resistance assay using RS toxin by: 1) developing a standardized method of toxin isolation, purification and delivery to rice plants in a consistent and reproducible manner, and 2) determining the relationship between toxin sensitivty and disease susceptibility in host plants.
Approach:
Prepare a standardized assay to isolate and quantify RS toxin for uniform resistance assays. Extend previous RS toxin work to include analytical methods for calculating toxin recovery we can make quantification absolute rahter than a relative estimate. Confirm the observed correlation between toxin sensitivity and disease susceptibility by surveying a large set of rice varieties with known SB disease ratings (MR-VS). Optimize our established infiltration method for rapid throughput and repeatability.
