Research Project: MOLECULAR CHARACTERIZATION OF THE TSN1 LOCUS IN WHEAT

Objective:
To identify, isolate, and validate Tsn1 candidate genes using molecular methods for analyzing knockout mutants.

Approach:
A BAC contig spanning the Tsn1 gene will be assembled, and the segment cosegregating with Tsn1 will be annotated to identity candidate genes. Seeds of the hexaploid wheat varieties Kulm and ND495 will be treated with the chemical mutagen EMS and fast-neutrons, respectively. The mutagenized populations will be grown and screened for reaction to Ptr ToxA to identify putative tsn1 knockouts. Tsn1 candidate genes will be cloned and sequenced from Kulm and the Kulm tsn1 knockout mutants to identify base substitutions within the candidate genes. The ND495 fast-neutron induced mutants will be screened for the presence/absence of candidate genes identified in the BAC contig.
