Objective:
Exploid existing Cry3Bb1-tolerant colonies of Western Corn Rootworm (WCR) to learn as much as possible about potential mechanisms of survival and any potential fitness costs associated with the resistance. Develop replicated colonies from three wild-type sources that will be crossed reciprocally to non-diapausing Colony 0. Larvae will be evaluated for survival on Bt and isoline corn, for survival in an established dose-response assay on synthetic Cry3Bb, and for behavioral responses in a series of bioassays.
Approach:
Develop replicated colonies of WCR populations reared on isoline or Bt corn. Determine the broad mechanism(s) of how WCR larvae survive Bt corn. Determine the fitness consequences of being selected to survive Bt corn.
