KSHV is a newly discovered herpes virus present in AIDS-KS lesions. KSHV can be continuously cultured in E lymphocytes derived from AIDS-related body cavity lymphomas. Ongoing mapping and sequence studies demonstrate that this virus is a gamma-2 herpesvirus that possesses metabolic, structural and transforming genes typically found in other gamma-2 herpesviruses. The virus is difficult to culture in vivo but low-titered transmission to other cells has been achieved. Multiple independent studies have found KSHV in nearly all KS tissues examined to date, including AIDS-KS, classical KS, African endemic KS and post-transplant KS lesions. The virus can be detected in peripheral blood mononuclear cells from AIDS patients prior to onset of KS and serologic studies suggest that AIDS-KS patients have high specific immunofluorescence antibody titers against KSHV which are not present among HIV patients without KS. These studies strongly suggest that KSHV/HHV8 is central to the pathogenisis of KS and possibly other AIDS-related lymphoproliferative disorders. Whether or not this herpesvirus, like most human herpesviruses, is ubiquitous among humans remains to be determined.
