Research Project: DIVERSITY IN WILD AND CULTIVATED DAUCUS

1a.Objectives (from AD-416)
To evaluate diversity in wild carrot, land races, and Daucus species in Poland where these populations all co-occur; to estimate genetic relationships and gene flow among them. Special attention to a transposon will be given.


1b.Approach (from AD-416)
Collect wild Daucus seed and Polish land races of carrots; grow plants, collect DNA, and evaluate nuclear and mitochondrial markers including DcMaster transposon.


3.Progress Report
This report serves to document accomplishments under a specific cooperative agreement between ARS and Krakow Agricultural University. Additional details of research can be found in the parent project 3655-21000-037-00D.

To date, 16 Polish wild Daucus carota populations, representing the species diversity in Poland, were collected. Additionally, Polish breeding companies donated DNA samples representing their breeding stocks of cultivated carrot (total of 45 accessions). The collection was supplemented with foreign material, including OP cultivars from Germany, Bielarus, Armenia, China, India, and USA, as well as few wild D. carota subspecies of European origin. Currently, the materials are grown in the field in order to evaluate their morphological characteristics.

The collected D. carota accessions are being investigated in terms of their diversity, using the standard Random Amplified Polymorphic DNA (RAPD) molecular marker system, as well as a newly developed DcMaster Transposon Display (DcMTD) system based on the DcMaster transposon insertion polymorphism. Both systems indicated a high level of both inter- and intrapopulation diversity. Over 70% DcMTD amplicons were polymorphic. Project monitored by e-mail discussions of progress every 6-8 weeks.
