Research Project: INCREASING THE DIVERSITY OF EST'S FOR FRAGARIA

Location: Genetic Improvement of Fruits and Vegetables

Project Number: 1275-21000-180-01
Project Type: Reimbursable

Start Date: Jun 01, 2005
End Date: May 31, 2008

Objective:
Project objectives: 1. Construction of 5 separate cDNA libraries from plants treated with a variety of abiotic stresses. 2. Sequencing of resultant clones to result in 35,000 sequences (ESTs). 3. Assembly of the sequences into Tentative Consensus (TC) sequences, annotation of TC sequences, and maintenance of the data in the TIGR Gene Index Database. 4. Dissemination of the data via GenBank and Genome Database for the Rosaceae.

Approach:
Aseptic seedlings of Fragaria vesca will be treated with various abiotic stresses including heat, cold, drought, and salt. RNA from these seedlings will be used to construct cDNA libraries. Approximately 50,000 clones from the resulting libraries will be single pass sequenced to give approximately 35,000 ESTs. The ESTs will be assembled into TCs, annotated and maintained in a database at TIGR.
