Citation: Mauro Herrera, M., Meerow, A.W., Borrone, J.W., Kuhn, D.N., Schnell Ii, R.J. 2006. Ten informative markers developed from wrky loci in coconut (cocos nucifera). Molecular Ecology Notes. 6:904-906.

Interpretive Summary: Ten new informative genetic markers were developed for coconut (Cocos nucifera) from a class of genes known to help plants resist disease and other stresses. These markers were evaluated in fifty individual palms representing five coconut varieties. These markers are currently being evaluated in other varieties. They show great promise estimating the genetic diversity of this economically important species.

Technical Abstract: Coconut (Cocos nucifera) WRKY gene sequences containing single nucleotide polymorphisms (SNPs) and one microsatellite repeat were used to develop ten informative markers. These markers were evaluated in fifty genotypes representing five coconut cultivars. Amplification products encompassing SNPs were analyzed using single-strand conformation polymorphism (SSCP). The number of detected alleles ranged from two to four, and only one locus showed significant deviation from Hardy-Weinberg equilibrium. Two pairs of loci were in linkage disequilibrium in the test population. These markers are currently being evaluated in other cultivars to determine their value in estimating the genetic diversity of this species.
