BACKGROUND:In the clinic, microdialysis is typically conducted for short durations (4-8 hours) in resting subjects with rigid probes, making this technique of limited use for profiling the skin penetration of anti-microbial agents with a long terminal elimination half-life. This study reports on the tolerability and precision of 24-hour ambulatory microdialysis. METHODS: Microdialysis (flexible CMA60) probes were inserted in the subcutaneous tissue of both arms of 11 male subjects, who received 500mg oral ciprofloxacin. Probes were perfused with Ringers solution at 2ul/min. Dialysate collections were made every hour, with concurrent plasma sampling. Determination of ciprofloxacin concentrations in human plasma and dialysate was by HPLC using single quadrupole mass spectroscopy. RESULTS: Subjects tolerated the probes well over 24 hours, with no adverse events relating to local tolerance. Uncorrected (for protein binding or in vivo recovery) mean pharmacokinetic parameters [95% confidence interval] were: [table: see text] CONCLUSIONS: 24-hour ambulatory microdialysis is well tolerated, and is particularly suitable for characterising subcutaneous pharmacokinetics of agents with a long terminal elimination half-live.
