Antipneumococcal Activity of BMS 284756, a New des-F(6)-Quinolone, Compared with Ten Other Agents.

Standard microdilution MIC methodology was used to test the activity of BMS-284756, a new quinolone, compared with ciprofloxacin, levofloxacin, moxifloxacin, amoxicillin +/- clavulanate, cefuroxime, ceftriaxone, imipenem, azithromycin and clarithromycin against 197 pneumococci (96 penicillin-susceptible, 31 penicillin-intermediate and 70 penicillin-resistant). MIC[50]/[90]s (microg/ml) were [table: see text]. BMS-284756 yielded the lowest MICs of all quinolones tested against all pneumococci irrespective of their penicillin or macrolide susceptibility status, followed by moxifloxacin, levofloxacin and ciprofloxacin. b-Lactam MICs rose with those of penicillin. Imipenem and ceftriaxone had the lowest MICs of all b-lactams, followed by amoxicillin (with or without clavulanate) and cefuroxime. Macrolide MICs rose with those of penicillin and were mainly seen in penicillin intermediate and resistant strains. Pneumococcal BMS 284756 MICs are amongst the lowest seen with all quinolones tested thus far against this species.KEYWORDS: BMS-284756; MIC; Pneumococci
