Quantitative modified plaque forming assay.

OBJECTIVES: To develop sensitive and simple method for the evaluation of anti-HIV compounds and assessment of neutralizing antibody response against HIV. METHODS: A modified plaque forming assay described by Harada et al (1985) was used. The modification concerned mainly the concentration of the MT-4 cells. (3-3, 5 x 10(6) cells/ml). Further steps have been completed as described by Harada et al (1985). RESULTS: It was shown that poly-L-lysin (PLL) could be substituted by human diploid cells (HDC). On the monolayer formed by HDC the MT-4 cell suspension was added (I, 5-2 x 10(6) cells/ml). After 24-48 h MT-4 cells formed a good monolayer. The diluted virus preparation was added to each dish and incubated for 60 min in a CO2 incubator at 37 degrees C. After adsorption of the virus the cells were washed with phosphate-buffered saline and the agarose medium (RPMI-I640 and Eagle medium in equal volumes) was overlayed. On the 6th day of incubation in a CO2 incubator at 37 degrees C the monolayer was stained with 0.1% neutral red. Both methods (with PLL and HDC) yielded similar results. Moreover MT-4 cells adjusted much better to the monolayer formed by HDC than to PLL. This method was used for evaluation of anti-HIV compounds. CONCLUSIONS: The described method could have particular application in testing of antiviral drugs in vitro, for the assay of virus infectivity, for the virus cloning.
