Research Interests -- Functional Genomics and Microbial Physiology

Current research projects in Dr. Brown’s laboratory include characterizing the dynamics of stress responses and genetic regulation in Zymomonas mobilis ZM4. We have developed a whole genome microarray that will be used in conjugation with other approaches such as metabolomic profiling and mutagenesis to elucidate the mechanisms and regulation of stress tolerance in strain ZM4. Dr. Brown has established a laboratory for controlled cultivation at ORNL that will allow control of, and log data for culture pH, temperature, level, dissolved oxygen tension (DOT) and thermal mass flow. Dr. Brown and Dr. Phelps are preparing to interface a GC-MS system for online metabolite analysis, so check back for updates soon. The cultivation laboratory also contains aerobic and anaerobic Bioscreen C machines for growth and monitoring of 200 individual cultures at a time. Dr. Brown has recently fabricated several whole genome microarrays for Clostridium thermocellum 27405 (lead-PI, Dr. Jonathan Mielenz) and Desulfovibrio desulfuricans G20 (lead-PI, Dr. Tony Palumbo) and is presently analyzing transcriptomic and other data as part of a larger systems biology team at ORNL. Dr. Brown has also conducted integrated biochemical, genomics and proteomic studies on Shewanella oneidensis MR-1 (more details).
