Abstract: (CIT): The fission yeast cdc5 gene was identified in a genetic screen for mutants defective in cell cycle progression. Since that time, our laboratory and others have found that Cdc5p and its orthologs in other organisms including human are essential components of a subcomplex of the spliceosome termed the nineteen complex (NTC). Using Cdc5p as a target for tandem affinity purification, we have determined the composition (by mass spectrometry) and structure (by cryo-electron microscopy) of the assembled spliceosome.
